Job description: Project description: The performance of topical formulations depends on their physicochemical properties and their interactions with the skin. Stratum corneum lipids are essential to skin barrier function. Their composition and organisation can be modified by applied formulations, potentially affecting the penetration and permeation of active substances. Understanding these interactions is necessary to improve the assessment of cutaneous products and the interpretation of in vitro release and permeation tests. This doctoral project, part of the European guideSkin doctoral network, aims to characterise formulation-induced changes in skin lipids and establish their relationships with barrier function and permeation. The research will be conducted at the Faculty of Pharmacy, Université Paris-Saclay, in collaboration with LVMH Recherche and RiverD International B.V. The project will combine in vivo confocal Raman spectroscopy, Fourier-transform infrared spectroscopy, liquid chromatography–mass spectrometry-based lipidomics, line-field confocal optical coherence tomography and skin biometric measurements. These complementary methods will investigate molecular composition, lipid organisation, skin microstructure and functional properties before and after formulation application. Biological variability, including age, sex and ethnicity, will be considered in study design and data analysis. Parallel in vitro release testing (IVRT) and in vivo skin permeation testing (IVPT) will identify situations in which formulation-induced modification of the skin barrier contributes to differences between release and permeation outcomes. Chemometric and multivariate statistical methods will be used to identify informative molecular and functional descriptors and relate them to formulation performance. Expected outcomes include optimised experimental protocols, a multimodal dataset describing formulation–skin lipid interactions, and descriptors supporting the network’s predictive modelling activities. The findings will contribute to a mechanistic understanding of topical formulation performance and inform approaches relevant to the assessment of cutaneous product equivalence. The doctoral training includes industrial secondments of 15 months at LVMH Recherche in France and 3 months at RiverD in the Netherlands.
